Simultaneous Quantitative and Qualitative Identification of Transformation Products of Triamcinolone Acetonide by Ozonation Using Liquid Chromatography (LC) Hyphenated with Quadrupole Time-of-flight Mass Spectrometry (Q-TOF-MS)

ABSTRACT

As water demand around the world increases partially due to climate change, securing potable water is increasingly more crucial. To this end, numerous water treatments, such as oxidation and physical separation with various water resources, such as river water and seawater, are considered. Among the treatment technologies, ozone oxidation is widely used for disinfection and attenuation of trace organic compounds, such as pharmaceutical and hormone. However, it inherently produces transformation products (TP). Hence, it is important to scrutinize the transformation products and their toxicity.

In this study, liquid chromatography (LC, Agilent 1290 Infinity Binary LC System) hyphenated with quadrupole time-of-flight (Q-TOF) mass spectrometry (MS) (Agilent 6540 Ultra High Definition Accurate Mass Q-TOF-MS) was utilized to identify transformation products of triamcinolone acetonide (TA), a glucocorticoid compound. Automatic MS/MS platform in the given equipment provides capability of simultaneous qualitative and quantitative analysis of TPs of TA. To be specific, automatic MS/MS triggers MS/MS mode from MS scan mode if ions are detected above a threshold abundance. The obtained MS/MS data were analyzed using Agilent MassHunter Workstation Software Qualitative analysis in order to obtain formula and abundances of TPs. The obtained MS/MS spectra were further analyzed using Agilent MassHunter Molecular Structure Correlator (MSC) in order to identify the structures of TPS of TA. As a result, the formula and structures of seven TPs by ozone oxidation were analyzed from a single injection. This automatic MS/MS platform can help expedite quantitative and structural analysis of not only transformation products by oxidation processes, but also other transformation products such as metabolites of biological processes.
